[Protocol - Washing Labtek Chambers for Single-Molecule Imaging]
for 8-well LabTek dishes
Remove the lids, arrange the Labtek dishes in a beaker, make sure the coverglass is vertical (as ultrasound is standing wave).
Add 1M KOH (Sigma-Aldrich) carefully into the beaker and make sure all the small wells are full of liquid (IMPORTANT!!!).
Sonicate the dishes for 30min to remove "residue" and add -OH to the SiO2.
Rinsed with large amounts of ddH2O (MilliQ) 4~5 times;
Sonicate coverslip in ddH2O (MilliQ) again for 10min to remove the KOH.
Briefly remove the water in the wells and air-dry overnight, under the shelf to avoid dust.
Check the autofluo dots on the surface with TIRF. (should be less than 20 dots per field of view)
Ready to use.
